Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251342 control chromosomes (gnomAD). c.155C>T has been observed in at least one individual affected with Medium Chain Acyl-CoA Dehydrogenase Deficiency (Maier_2005). At least two publications report experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 30%-50% of normal activity (Maier_2009, Jank_2014). The following publications have been ascertained in the context of this evaluation (PMID: 32778825, 19673949, 24718418, 19224950, 15832312). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
