NM_153252.5(BRWD3):c.2327A>T (p.Asn776Ile) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The N776I variant in the BRWD3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The N776I variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The N776I variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Asparagine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret N776I as a variant of uncertain significance.
