This variant lies in the ANK3 gene (transcript NM_020987.5) at coding-DNA position 6895, where G is replaced by A; at the protein level this means replaces alanine at residue 2299 with threonine — a missense variant. Submitter rationale: The A2299T variant in the ANK3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A2299T variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The A2299T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret A2299T as a variant of uncertain significance.
