This variant lies in the DSE gene (transcript NM_013352.4) at 6 bases into the intron immediately before 53 bases upstream of the translation start (5' untranslated region), where A is replaced by G. Submitter rationale: Variant summary: DSE c.-53-6A>G is located in the untranslated mRNA region upstream of the initiation codon. The variant alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predicts no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 245424 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.-53-6A>G in individuals affected with Ehlers-Danlos syndrome, musculocontractural type 2 and no experimental evidence demonstrating its impact on protein function have been reported.
