Likely pathogenic for Recessive dystrophic epidermolysis bullosa — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000094.4(COL7A1):c.7949G>A (p.Gly2650Asp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL7A1 c.7949G>A (p.Gly2650Asp) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. This variant disrupts the triple helix domain of COL7A1. Glycine residues within the Gly-Xaa-Yaa repeats of the triple helix domain are required for the structure and stability of fibrillar collagens (PMID: 7695699, 8218237, 19344236). The variant was absent in 251248 control chromosomes. c.7949G>A has been observed in at least one homozygous individual affected with Dystrophic Epidermolysis Bullosa, Recessive (e.g. Vahidnezhad_2017). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
