Uncertain significance for Catecholaminergic polymorphic ventricular tachycardia 5; Inducible ventricular tachycardia — the classification assigned by Clinical Genomics Laboratory, Stanford Medicine to NM_006073.4(TRDN):c.1484A>G (p.Lys495Arg), citing ACMG Guidelines, 2015: The p.Lys495Arg variant in the TRDN gene has not been previously reported in association with disease. This variant was absent from large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/). Computational tools predict that this variant does not impact protein function; however, the accuracy of in silico algorithms is limited. These data were assessed using the ACMG/AMP variant interpretation guidelines. In summary, the significance of the p.Lys495Arg variant is uncertain.
