Likely pathogenic for Ehlers-Danlos syndrome, musculocontractural type 1 — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NM_130468.4(CHST14):c.870_871delinsT (p.Val291fs), citing ACMG Guidelines, 2015: A novel frameshift variant, c.870_871delinsT in CHST14 was identified in a homozygous state in the proband. This variant was present in a heterozygous state in her parents. The above-mentioned variant is not present in the gnomAD population database and our in-house exome data of 3145 individuals. In silico tool Mutation Taster predicts the variant to be disease-causing, resulting in the formation of truncating or non-functional protein. Clinical findings observed in the proband are suggestive of Ehlers-Danlos syndrome, musculocontractural type 1. Thus, the above-mentioned findings confirm the diagnosis of Ehlers-Danlos syndrome, musculocontractural type 1 in Keerthana.
