Likely pathogenic for Tooth agenesis, selective, 7; Tooth agenesis — the classification assigned by Center of Excellence in Genomics and Precision Dentistry, Faculty of Dentistry, Chulalongkorn University to NM_002336.3(LRP6):c.1231G>T (p.Asp411Tyr), citing ACMG Guidelines, 2015: The de novo missense variant c.1231G>T (p. Asp411Tyr) in LRP6 gene was identified in a patient with nonsyndromic tooth agenesis, and was absent from population database. This variant is located in the LDL recepter class B domain and predicted to be pathogenic by multiple prediction tools. The missense variant in this domain have been reported in the patients with familal tooth agenesis (PMID: 34834569). It was classified as likely pathogenic according to ACMG standard guidelines for variant classification.
