Uncertain significance — the classification assigned by GeneDx to NM_173551.5(ANKS6):c.2593C>T (p.Pro865Ser), citing GeneDx Variant Classification (06012015): The P865S variant in the ANKS6 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. Although not present in the homozygous state, the P865S variant was observed on 97 of 7922 alleles (1.2%) from individuals of East Asian background, in the Exome Aggregation Consortium (ExAC) data set, indicating it may be a rare variant in this population. The P865S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution also occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P865S as a variant of uncertain significance.
