Likely oncogenic for Meningioma — the classification assigned by Dr. Guy Rouleau's laboratory, McGill University to NM_032271.3(TRAF7):c.1922G>A (p.Arg641His), citing ClinGen/CGC/VICC Guidelines for Oncogenicity, 2022: This missense variant, located at position 641 in the TRAF7 gene, results in the substitution of Arginine (R), a basic amino acid, with Histidine (H), another basic amino acid featuring an imidazole side chain. This somatic variant was identified in a paired tumor-blood sequencing study of meningiomas. It has been reported in meningiomas (PMIDs: 30546952, 31963394, 33407897, 35299730, 35984495). However, it has not been documented in population databases (gnomAD v2.1.1: no frequency).
