Uncertain significance for Meningioma — the classification assigned by Dr. Guy Rouleau's laboratory, McGill University to NM_032271.3(TRAF7):c.1730A>G (p.Tyr577Cys), citing ClinGen/CGC/VICC Guidelines for Oncogenicity, 2022. This variant lies in the TRAF7 gene (transcript NM_032271.3) at coding-DNA position 1730, where A is replaced by G; at the protein level this means replaces tyrosine at residue 577 with cysteine — a missense variant. Submitter rationale: This missense variant, located at position 577 in the TRAF7 gene, results in the substitution of Tyrosine (Y), an aromatic amino acid, with Cysteine (C), a sulfur-containing amino acid. This somatic variant was identified in a paired tumor-blood sequencing study of meningiomas. It has been reported in Well-differentiated papillary mesothelioma (PMID: 30171198). However, it has not been reported in population databases (gnomAD v2.1.1: no frequency). Due to insufficient evidence, the clinical significance of this variant remains unknown
