NM_198253.3(TERT):c.1324del (p.Asp442fs) was classified as Likely oncogenic for Meningioma by Dr. Guy Rouleau's laboratory, McGill University, citing ClinGen/CGC/VICC Guidelines for Oncogenicity, 2022: This frameshift variant generates a premature translational stop signal (p.Asp442Ter) in the TERT gene, which is expected to result in an absent or disrupted protein product. Loss-of-function variants in TERT are well-established as pathogenic (PMIDs: 16247010, 17460043, 32923861). This somatic variant was identified in a paired tumor-blood sequencing study of meningiomas. However, it has not been reported in population databases (gnomAD v2.1.1: no frequency).
