Oncogenic for Meningioma — the classification assigned by Dr. Guy Rouleau's laboratory, McGill University to NM_000268.4(NF2):c.1030C>T (p.Gln344Ter), citing ClinGen/CGC/VICC Guidelines for Oncogenicity, 2022: This nonsense variant generates a premature translational stop signal (p.Gln344Ter) in the NF2 gene, which is expected to result in an absent or disrupted protein product. Loss-of-function variants in NF2 are well-established as pathogenic (PMIDs: 8755919, 9643284, 16983642). This somatic variant was identified in a paired tumor-blood sequencing study of meningiomas. It has also been reported in three cases of spinal meningiomas (PMID: 32724039). However, it has not been reported in population databases (gnomAD v2.1.1: no frequency).
