NM_002474.3(MYH11):c.5100G>C (p.Glu1700Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1700D variant (also known as c.5100G>C), located in coding exon 35 of the MYH11 gene, results from a G to C substitution at nucleotide position 5100. The glutamic acid at codon 1700 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
