Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000017.10:g.(17125976_17127235)_(17127458_17129489)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant involves the duplication of exon 6 in the FLCN gene. A presumed nomenclature of c.(396+1_397-1)_(618+1_619-1)dup has been designated for the purposes of this classification. It is assumed to be a tandem duplication in direct orientation (PMIDs: 25640679, 30054569). This Copy Number Variant (CNV) is predicted to result in an in-frame duplication within this gene. The variant was absent in 120750 control chromosomes in the gnomAD database (Structural Variants v4.1 dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(396+1_397-1)_(618+1_619-1)dup in individuals affected with Birt-Hogg-Dube Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1449742). Based on the evidence outlined above, the variant was classified as uncertain significance.
