Likely pathogenic for Thyroid dyshormonogenesis 6 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001363711.2(DUOX2):c.534G>T (p.Trp178Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DUOX2 c.534G>T (p.Trp178Cys) results in a non-conservative amino acid change located in the peroxidase domain (IPR034821) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 141452 control chromosomes (gnomAD). c.534G>T has been reported in the literature in at least two compound heterozygous individuals affected with Thyroid Dyshormonogenesis 6 (e.g. Tan_2016, Sun_2018), who carried a (likely) pathogenic variant on the other allele. These data indicate that the variant may be associated with disease. At least one publication reported experimental evidence evaluating an impact on protein function and demonstrated that the variant results in about 20% of normal activity (Sun_2021). The following publications have been ascertained in the context of this evaluation (PMID: 29650690, 34564849, 27557340). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as likely pathogenic.
