NM_001278716.2(FBXL4):c.-73+1G>T was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: FBXL4 c.-73+1G>T is located in the untranslated mRNA region upstream of the initiation codon. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The FBXL4 canonical start codon is located in exon 3, downstream of the variant. The variant was absent in 31406 control chromosomes. To our knowledge, no occurrence of c.-73+1G>T in individuals affected with Leigh Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
