This variant lies in the COL4A4 gene (transcript NM_000092.5) at coding-DNA position 2986, where G is replaced by C; at the protein level this means replaces glycine at residue 996 with arginine — a missense variant. Submitter rationale: Variant summary: COL4A4 c.2986G>C (p.Gly996Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249466 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2986G>C has not been reported in the literature. However, another variant c.2986G>A (p.Gly996Arg) has been observed in individuals with hematuria/proteinuria and Alport-related symptoms (Kovcs_2016, Li_2020). These reports do not provide unequivocal conclusions about association of this variant with Alport Syndrome, Autosomal Recessive.
