This variant lies in the TGM1 gene (transcript NM_000359.3) at coding-DNA position 1097, where T is replaced by C; at the protein level this means replaces leucine at residue 366 with proline — a missense variant. Submitter rationale: Variant summary: TGM1 c.1097T>C (p.Leu366Pro) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251260 control chromosomes. c.1097T>C has been observed in at-least two compound heterozygous individual(s) affected with Lamellar Ichthyosis (example: Cucuzza_2018, Herman_2009). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
