Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 200930 control chromosomes. c.779C>T has been reported in the literature in at least two compound heterozygous individuals affected with Mucopolysaccharidosis Type 1 (e.g. Matte_2003, Thomas_2021). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function in vitro and found that the variant effect results in <10% of WT activity (Matte_2003).
