This variant lies in the CHEK2 gene (transcript NM_007194.4) at coding-DNA position 106, deleting one base; at the protein level this means shifts the reading frame starting at glutamine residue 36, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Variant summary: CHEK2 c.106delC (p.Gln36ArgfsX25) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 250608 control chromosomes. c.106delC has been reported in the literature as found in an individual from a study cohort of CHEK2 germline mutation carriers (Borde_2020). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
