This variant lies in the TYRP1 gene (transcript NM_000550.3) at coding-DNA position 1012, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 338 with lysine — a missense variant. Submitter rationale: Variant summary: TYRP1 c.1012G>A (p.Glu338Lys) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 250832 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance (gnomAD). c.1012G>A has been reported in the literature in one individual affected with cutaneous melanoma (Nathan_2019). The report does not provide unequivocal conclusions about association of the variant with Oculocutaneous albinism type 3. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
