This variant lies in the ATP7B gene (transcript NM_000053.4) at coding-DNA position 2278, where C is replaced by T; at the protein level this means replaces proline at residue 760 with serine — a missense variant. Submitter rationale: Variant summary: ATP7B c.2278C>T (p.Pro760Ser) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249582 control chromosomes (gnomAD). c.2278C>T has been reported in the literature in individuals affected with Wilson Disease (e.g. Liu_2022, Zhang_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
