A presumed nomenclature of c.(?_-66)_(208+1_209-1)del has been designated for the purposes of this classification. The exact breakpoint at the 5' end of this variant is unknown, therefore this deletion may extend upstream of the annotated region of this gene. Although the exact breakpoints of this deletion are not known, it is predicted to remove the initiation codon and result in an absence of protein or a truncation of the encoded protein due to translation initiation at a downstream site. The variant allele was found at a frequency of 9.2e-05 in 21694 control chromosomes (2/21694 alleles in the gnomAD structural variants dataset). Various deletion variants encompassing exon 1 of the SAMHD1 gene have been reported in the literature in multiple homozygous and compound heterozygous individuals affected with Aicardi-Goutieres syndrome 5 (Piccoli_2021, Ramesh_2010, Leshinsky-Silver_2011, Rice_2013) and is considered an Ashkenazi Jewish founder mutation (Rice_2013). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
