Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002480.3(PPP1R12A):c.2667-5T>C, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PPP1R12A c.2667-5T>C alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant weakens a 3' acceptor site. One predict the variant no significant impact on splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.3e-07 in 1581436 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2667-5T>C in individuals affected with Genitourinary And/or Brain Malformation Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
