Submitter rationale: The CDKN2A locus encodes two different gene products, p16INK4a and p14ARF (https://www.ncbi.nlm.nih.gov/books/NBK7030/). This variant causes an A to C nucleotide substitution at the first coding nucleotide of intron 1 of the CDKN2A (p16INK4A) mRNA gene, altering the translation initiation codon. However, another in-frame methionine is located at codon 9, which may be able to rescue translation initiation. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with CDKN2A-related disorders in the literature.
