Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_001035.3(RYR2):c.3595G>A (p.Asp1199Asn), citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 3595, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 1199 with asparagine — a missense variant. Submitter rationale: This missense variant replaces aspartic acid with asparagine at codon 1199 of the RYR2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two related individuals affected with familial intracranial aneurysm and in an unaffected individual from the family (PMID: 34668355). This variant has been identified in 16/247842 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
