Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.1067A>G (p.Lys356Arg), citing Ambry Variant Classification Scheme 2023: The p.K356R variant (also known as c.1067A>G), located in coding exon 10 of the PMS2 gene, results from an A to G substitution at nucleotide position 1067. The lysine at codon 356 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
