NM_000179.3(MSH6):c.3284dup (p.His1096fs) was classified as Likely pathogenic for Lynch syndrome 5 by Servicio Canario de Salud, Hospital Universitario Nuestra Sra. de Candelaria, citing ACMG Guidelines, 2015: The c.3284dup; p.(His1096ProfsTer12) MSH6 variant has been reported in our laboratory in a 58-year-old male patient with a clinical diagnosis of melanoma and urothelial and lung cancer. Family history: father colon cancer. This variant has never been reported in MSH6 related-disorders. This variant was absent from large population studies (gnomAD no frequency). In summary, the available evidence for c.3284dup; p.(His1096ProfsTer12) MSH6 variant meets our criteria to be classified as Likely Pathogenic based upon its absence from controls and the clinical correlation in this patient´s phenotype.
