This variant lies in the XPC gene (transcript NM_004628.5) at coding-DNA position 398, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 133 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The novel stop-gain variant c.398G>A in exon 3 of XPC, was observed in heterozygous state in the father. This variant is not observed in homozygous and/or heterozygous state in gnomAD (v4.1.0) population database and our in-house database of 3464 individuals.
