Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.2477A>T (p.Glu826Val), citing Ambry Variant Classification Scheme 2023: The p.E826V variant (also known as c.2477A>T), located in coding exon 15 of the PMS2 gene, results from an A to T substitution at nucleotide position 2477. The glutamic acid at codon 826 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
