NM_000535.7(PMS2):c.336C>G (p.Ser112Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S112R variant (also known as c.336C>G), located in coding exon 4 of the PMS2 gene, results from a C to G substitution at nucleotide position 336. The serine at codon 112 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
