NM_000535.7(PMS2):c.2455G>T (p.Gly819Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G819W variant (also known as c.2455G>T), located in coding exon 15 of the PMS2 gene, results from a G to T substitution at nucleotide position 2455. The glycine at codon 819 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
