Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.453C>G (p.Phe151Leu), citing Ambry Variant Classification Scheme 2023: The p.F151L variant (also known as c.453C>G), located in coding exon 4 of the PLOD1 gene, results from a C to G substitution at nucleotide position 453. The phenylalanine at codon 151 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
