Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.986A>G (p.His329Arg), citing Ambry Variant Classification Scheme 2023: The p.H329R variant (also known as c.986A>G), located in coding exon 10 of the PLOD1 gene, results from an A to G substitution at nucleotide position 986. The histidine at codon 329 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
