Uncertain significance — the classification assigned by Ambry Genetics to NM_003628.6(PKP4):c.73G>A (p.Gly25Ser), citing Ambry Variant Classification Scheme 2023: The p.G25S variant (also known as c.73G>A), located in coding exon 1 of the PKP4 gene, results from a G to A substitution at nucleotide position 73. The glycine at codon 25 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
