This variant lies in the KAT6A gene (transcript NM_006766.5) at coding-DNA position 5878, where A is replaced by C; at the protein level this means replaces serine at residue 1960 with arginine — a missense variant. Submitter rationale: The S1960R variant in the KAT6A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S1960R variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S1960R variant is a semi-conservative amino acid substitution, which may impact secondary protein structureas these residues differ in some properties. This substitution occurs at a position that is conserved inmammals. However, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
