NM_022436.3(ABCG5):c.1101A>T (p.Lys367Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K367N variant (also known as c.1101A>T), located in coding exon 8 of the ABCG5 gene, results from an A to T substitution at nucleotide position 1101. The lysine at codon 367 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
