NM_016734.3(PAX5):c.640C>T (p.Leu214Phe) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L214F variant (also known as c.640C>T), located in coding exon 6 of the PAX5 gene, results from a C to T substitution at nucleotide position 640. The leucine at codon 214 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
