Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.*175T>A, citing Ambry Variant Classification Scheme 2023: The p.N663K variant (also known as c.1989T>A), located in coding exon 11 of the PALLD gene, results from a T to A substitution at nucleotide position 1989. The asparagine at codon 663 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
