NM_024675.4(PALB2):c.790C>G (p.His264Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H264D variant (also known as c.790C>G), located in coding exon 4 of the PALB2 gene, results from a C to G substitution at nucleotide position 790. The histidine at codon 264 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
