Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.10805G>A (p.Cys3602Tyr), citing Ambry Variant Classification Scheme 2023: The p.C3173Y variant (also known as c.9518G>A), located in coding exon 35 of the OBSCN gene, results from a G to A substitution at nucleotide position 9518. The cysteine at codon 3173 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
