Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.16856G>A (p.Arg5619His), citing Ambry Variant Classification Scheme 2023: The p.R4662H variant (also known as c.13985G>A), located in coding exon 53 of the OBSCN gene, results from a G to A substitution at nucleotide position 13985. The arginine at codon 4662 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
