Uncertain significance — the classification assigned by GeneDx to NM_173551.5(ANKS6):c.407A>C (p.Asp136Ala), citing GeneDx Variant Classification (06012015): The D136A variant in the ANKS6 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The D136A variant was not observed in approximately 6200 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The D136A variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Aspartic Acid are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret D136A as a variant of uncertain significance.
