Uncertain significance — the classification assigned by GeneDx to NM_001853.4(COL9A3):c.1109G>A (p.Gly370Glu), citing GeneDx Variant Classification (06012015). This variant lies in the COL9A3 gene (transcript NM_001853.4) at coding-DNA position 1109, where G is replaced by A; at the protein level this means replaces glycine at residue 370 with glutamic acid — a missense variant. Submitter rationale: The G370E variant in the COL9A3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G370E variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G370E variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G370E as a variant of uncertain significance.
