Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_052876.4(NACC1):c.271A>C (p.Ser91Arg), citing Ambry Variant Classification Scheme 2023: The c.271A>C (p.S91R) alteration is located in exon 2 (coding exon 1) of the NACC1 gene. This alteration results from a A to C substitution at nucleotide position 271, causing the serine (S) at amino acid position 91 to be replaced by an arginine (R). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is well conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
