Likely pathogenic — the classification assigned by GeneDx to NM_001079866.2(BCS1L):c.725C>G (p.Ala242Gly), citing GeneDx Variant Classification (06012015): The A242G variant in the BCS1L gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A242G variant is not observed in large population cohorts (Lek et al., 2016). The A242G variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. However, in-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret A242G as a likely pathogenic variant.
