This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 1483, where A is replaced by T; at the protein level this means replaces threonine at residue 495 with serine — a missense variant. Submitter rationale: The p.T495S variant (also known as c.1483A>T), located in coding exon 9 of the MSH2 gene, results from an A to T substitution at nucleotide position 1483. The threonine at codon 495 is replaced by serine, an amino acid with similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was determined to be functionally neutral (Jia X et al.
