Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.1920_1921del (p.Leu641fs), citing Ambry Variant Classification Scheme 2023: The c.1920_1921delCC pathogenic mutation, located in coding exon 17 of the MLH1 gene, results from a deletion of two nucleotides at nucleotide positions 1920 to 1921, causing a translational frameshift with a predicted alternate stop codon (p.L641Sfs*3). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
