NM_001354604.2(MITF):c.1555A>G (p.Met519Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M412V variant (also known as c.1234A>G), located in coding exon 9 of the MITF gene, results from an A to G substitution at nucleotide position 1234. The methionine at codon 412 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
